Nicotinamide adenine dinucleotide (NAD+) is the most important metabolite (no one) for maintaining cell viability. As people getting old, NAD+ levels will continue to decline and cell viability will enter the downward channel and lead to individual aging. Nicotinamide mononucleotide (NMN) is the direct precursor of NAD+. Supplementing NMN can effectively improve NAD+. There is a negatively charged phosphate group on the metabolite of NMN. This molecular structure prevents NMN from effectively passing through the cell membrane. So, the question is how does NMN achieve transmembrane transport into the cell? If NMN cannot effectively enter the cell, then the effect of NAD+ produced by NMN will be compromised.

Shin-ichiro Imai, a Japanese scientist from the University of Washington in the United States, discovered a transporter protein Slc12a8 on the cell membrane, which helps NMN enter the cell across the membrane.

Scientists found that:

1. The Slc12a8 gene encodes a specific NMN cell membrane transporter protein, which helps NMN enter the cell;

2. Slc12a8 specifically transports NMN and has no effect on other precursors such as nicotinamide nucleotide (NR) and nicotinamide (NAM);

3. The transporter Slc12a8 is highly expressed in the small intestine and pancreas, and moderately expressed in the liver and white adipose tissue;

4. In the aging mouse model, oral administration of NMN, compared with young mice, will cause the plasma NMN level of old mice increased by more times, and the NAD+ level in the small intestine returned to the level close to that of young mice;
