RGD-全氟化碳纳米乳MRI显影剂的制备技术
1.一种RGD-全氟化碳纳米乳MRI显影剂的制备方法,其特征在于,包括如下步骤:
(1)纳米乳的制备:先将占磷酸盐缓冲液4wt%的磷脂和占磷酸盐缓冲液0.5wt%的普朗尼克F-68溶解在1ml磷酸盐缓冲液(PBS)中,然后将占磷酸盐缓冲液10wt%-20wt%的全氟溴辛烷(PFOB)加入到上述缓冲液中,并在超声仪中处理5-10分钟,再用试管振荡器振荡10-15分钟,得粗乳液;之后将制备的粗乳液用去离子水稀释20倍,再通过微孔滤膜挤制2次,得到纳米乳记为NPs;
(2)RGD-纳米乳的制备:将胆固醇-PEG2000-RGD加入到步骤(1)制备的纳米乳液中,然后置于摇床上以300rpm的速度振荡1小时得到靶向纳米乳NPs-RGD;
(3)荧光标记的RGD纳米乳的制备:向步骤(2)制备的靶向纳米乳中加入浓度为5mg/mL的荧光染料DiI,然后置于摇床上以300rpm的速度振荡1小时,得到DiI标记的RGD-纳米乳。
2.根据权利要求1所述的制备方法,其特征在于,所述胆固醇-PEG2000-RGD的制备方法为:将胆固醇-PEG2000-Mal与RGDyc按摩尔比10:1-1:1加入1ml PBS中,于室温下置于摇床上以300rpm的速度振荡20-24小时,然后用透析袋在超纯水中透析去除未反应的RGDyc分子,经冷冻干燥机冻干得到靶向分子胆固醇-PEG2000-RGD。
3.根据权利要求1所述的制备方法,其特征在于,所述磷脂为大豆卵磷脂(Lipoid S75)。
4.根据权利要求1所述的制备方法,其特征在于,所述磷酸盐缓冲液的浓度为5mM,pH为7.4。
5.根据权利要求1所述的制备方法,其特征在于,所述微孔滤膜的孔径为0.22μm。
6.根据权利要求1-5任一项所述的制备方法,其特征在于,所述胆固醇-PEG2000-RGD的加入量与磷脂的摩尔比为1:20。
7.根据权利要求1-6任一项所述的制备方法,其特征在于,步骤(1)中磷脂与F-68的质量比为8:1。
8.根据权利要求1-7任一项所述的制备方法,其特征在于,所述磷脂与全氟溴辛烷(PFOB)的质量比为1:2.5-1:5。
9.根据权利要求1-7任一项所述方法制备得到的RGD-全氟化碳纳米乳MRI显影剂。
