反相液相色谱法测定老鼠以及酵母中的同位素代谢物

用两种不同的液相分离(亲水性和疏水性)和质谱联用的技术,鉴定老鼠以及酵母中的同位素代谢物。设定了较佳的鉴定参数,并且对正常酵母和氮饥饿的酵母样品,进行同位素代谢组学鉴定。

反相液相色谱法(reverse-phase liquid chromatography,RPLC)是代谢组学中较常见和实用的一种LC分离方法。但是缺点是对于强极性物质保留较差。亲水相互作用液相色谱(hydrophilic interaction liquid chromatography,HILIC)是近年来色谱领域研究的热点之一。

它通过采用强极性度固定相,并且结合高比例有机相和低比例水相作为流动相。此前文章报道的RPLC和HILIC是主要集中在特征峰的数目方面,还没有文章系统比较这两种方法在代谢组学上的鉴定和分析。

系统地比较了8种不同条件的RPLC-MS和HILIC-MS方法在代谢组学鉴定上的不同,为代谢组学的鉴定提供新方法。
